Abstract

Abstract A vaccine against a DNA virus, for example, Herpes simplex virus, is prepared by incubating a cell sample which has been infected with the virus releasing the nuclei in the infected cell material from the cytoplasmic fraction of that material, chemically fixing the polypeptide chain in the cytoplasmic fraction, and forming a precipitate which includes the virus antigens in the cytoplasmic fraction, the precipitate providing the active constituent of the vaccine. A characteristic strain of the virus is preferred, and transfers its characteristics to the vaccine so that vaccinated subjects may subsequently be distinguished from infected subjects.

Claims Analysis

Total Claims: 24
Claim 1
Independent Claim 67 words
A method of preparing a vaccine against a DNA virus includes the steps of incubating a cell sample which has been infected with the viurus, and forming a precipitate which includes the virus antigens in the infected cell sample, the precipitate providing the active constituent of the vaccine, characterised in that before forming said precipitate the polypeptide chains in said cytoplasmic fractions are fixed by chemical reaction.
Claim 2
Independent Claim 67 words
A method of preparing a vaccine against a DNA virus includes the steps of incubating a cell sample which has been infected with the viurus, and forming a precipitate which includes the virus antigens in the infected cell sample, the precipitate providing the active constituent of the vaccine, characterised in that before forming said precipitate the polypeptide chains in said cytoplasmic fractions are fixed by chemical reaction.
Claim 3
Dependent Claim 35 words
A method as claimed in claim 1 which includes the step of releasing the nuclei of the infected cell material from the cytoplasmic fraction of that material, said precipitate being formed from said cytoplasmic fraction.
Claim 4
Dependent Claim 26 words
A method as claimed in claim 2 in which fixing of the polypeptide chains is effected by addition of formaldehyde or glutaraldehyde to said cytoplasmic fraction.
Claim 5
Dependent Claim 23 words
A method as claimed in claim 2 which includes the steps of separating and discarding the released nuclei, and retaining the cytoplasmic fraction.
Claim 6
Dependent Claim 36 words
A method as claimed in claim 2 in which said precipitate is formed by steps which include reacting said cytoplasmic fraction with antibodies to the proteins of said virus, and separating the antigen/antibody precipitate so formed.
Claim 7
Independent Claim 43 words
A method as claimed in claim .2 in which said precipitate is formed by steps which include reacting said cytoplasmic fraction with antibodies to the proteins in said cell sample, separating and discarding the antigen/antibody precipitate so formed, and precipitating the remaining proteins.
Claim 8
Dependent Claim 33 words
A method as claimed in claim 2, which includes the steps of cultivating said cells in a serum, and subsequently subjecting the cultivated cells to serum deprivation prior to infection with said virus.
Claim 9
Dependent Claim 39 words
A method as claimed in claim 7 in which said precipitate is formed by steps which include the addition to said cytoplasmic fraction of an organic solvent which reacts with water in preference to the proteins in said sample.
Claim 10
Dependent Claim 23 words
A method as claimed in claim 8 in which said organic solvent is selected from the group which includes acetone, ethanol and methanol.
Claim 11
Dependent Claim 24 words
A method as claimed in claim 8 or claim 9 in which 'the temperature of said solvent is between 0°C and - 300 C.
Claim 12
Independent Claim 23 words
A method as claimed in any one of claims 5, 6 or 7 which includes the addition of said precipitate to aluminium hydroxide.
Claim 13
Dependent Claim 23 words
A method as claimed in claim 7, in which said precipitate is formed by the addition of aluminium hydroxide to said cytoplasmic fraction.
Claim 14
Dependent Claim 16 words
A vaccine against DNA virus, when prepared by a method as claimed in any preceding claim.
Claim 15
Dependent Claim 24 words
A vaccine against Herpes simplex virus, characterized by the presence of proteins of the molecular weights substantially as indicated in Figures 1 and 2.
Claim 16
Dependent Claim 24 words
A vaccine against Herpes simplex virus, characterized by the presence of proteins of the molecular weights substantially as indicated in Figures 1 and 2.
Claim 17
Dependent Claim 20 words
A vaccine against Herpes simplex virus, characterized by the absence of proteins from serum used in preparation of the vaccine.
Claim 18
Dependent Claim 20 words
A vaccine against Herpes simplex virus, characterized by the absence of proteins from serum used in preparation of the vaccine.
Claim 19
Dependent Claim 21 words
A vaccine as claimed in claim 15, characterized by the absence of proteins from cells used in preparation of the vaccine.
Claim 20
Independent Claim 25 words
A method of protecting a subject against Herpes simplex virus, comprising administration of a vaccine as claimed in any of claims 13, 14 or 15.
Claim 21
Dependent Claim 24 words
A herpes simplex type 1 virus characterised by the presence of proteins of the undecular weights substantially as indicated in Figures 1 and 2.
