Abstract

Abstract A diffusional drug delivery device is described which can provide for stability of the adhesive and system components, elimination of the initial burst of drug and hence irritation, and to provide for delayed onset of therapeutic effect along with delivery of a therapeutic agent at an optimum rate. The therapeutic agent in a first form which is suitable for storage, and the anhydrous activating means are inert when in an anhydrous environment. Moisture activates the system whereby the activating means provides an acidic or basic solution and the first form of the therapeutic agent is converted to a second form which is suitable for absorption through the skin or mucosa.

Claims Analysis

Total Claims: 12
Claim 1
Independent Claim 154 words
A controlled release medical device for the delivery to a biological environment of at least one therapeutic agent in a predetermined delivery rate pattern; which device is characterised by a therapeutic agent capable of existing in a first form suitable for storage and in a second form suitable for adsorption through skin and mucosa, reservoir means containing said therapeutic agent in said first form and have a surface substantially impermeable to the therapeutic agent in its first form and permeable to said therapeutic agent in said second form, activating means containing an activating agent which in a first state is anhydrous and in a second state is hydrated, wherein in the environment of use the activating agent transmogrifies from the anhydrous to the hydrated state in which latter form it causes or allows the therapeutic agent to assume the said second form in which it can permeate the reservoir surface and enter the environment.
Claim 2
Dependent Claim 154 words
Claim 3
Dependent Claim 29 words
A device as claimed in claim 1 wherein the activating means is moistened by cutaneous liquids whereby said activating agent is changed from said first to said second state.
Claim 4
Dependent Claim 52 words
A device as claimed in claim 1 or claim 2 wherein said reservoir means is a matrix having a therapeutic agent dispersed throughout, said means being substantially free of activating agent, and said activating means is a matrix having an activating agent dispersed throughout, said means being substantially free of therapeutic agent.
Claim 5
Independent Claim 32 words
A device as claimed in any preceding claim further characterised by rate controlling means for controlling the rate at which said activating means becomes hydrated when placed at its environment of use.
Claim 6
Independent Claim 32 words
A device as claimed in any preceding claim further characterised by rate controlling means for controlling the rate at which said activating agent in its second state diffuses into said reservoir means.
Claim 7
Independent Claim 38 words
A device as claimed in any preceding claim wherein said reservoir means and said activating means comprise a single reservoir having a therapeutic agent and activating agent contained therein, in a dry blend or dispersed throughout a matrix.
Claim 8
Independent Claim 51 words
A device as claimed in any preceding claim wherein said reservoir means is comprised of a therapeutic agent in a non-aqueous medium, said means being substantially free of activating agent, and said activating means is a matrix having an activating agent dispersed throughout, said means being substantially free of therapeutic agent.
Claim 9
Independent Claim 25 words
A device as claimed in any preceding claim wherein said reservoir means is comprised of a first and a second therapeutic agent which are co-delivered.
Claim 10
Dependent Claim 47 words
A device as claimed in claim 8 wherein said first therapeutic agent is a non-salt drug and said second therapeutic agent in a first form is a salt form of a drug, and delivery of the second therapeutic agent is delayed until a change of state occurs.
Claim 11
Independent Claim 38 words
A device as claimed in any preceding claim wherein said reservoir means is comprised of a first and a second therapeutic agent, where delivery of the second therapeutic agent commences when delivery of the first therapeutic agent ceases.
Claim 12
Independent Claim 39 words
A device as claimed in any preceding claim wherein said therapeutic agent is selected from fluorouracil, barbitol, furosemide, albuterol, apomorphine, benzocaine, acetylsalicylic acid, scopolamine, clonidine, phenylpropanolamine, chlorpheniramine, pilocarpine, terbutaline, salbutamol, ephedrine, ergonovine, ergotamine, benztropine, nicotine, secoverine, propanolol and timolol.
