Abstract

Abstract Disclosed is a diagnostic test device which comprises a fluid permeable structural support member having a macroporous membrane, i.e., having pores greater than 0.2 µ in diameter on one side thereof and a microporous membrane, i e., having pores less than 0.2 µ in diameter on the other side. The device is structured so as to allow the free flow of fluid applied to the macroporous membrane through to the microporous membrane. The device is designed to facilitate the inclusion of incompatible reagents in a single test device.

Claims Analysis

Total Claims: 10
Claim 1
Independent Claim 27 words
A diagnostic test device comprising a fluid permeable structural support member having a macroporous membrane on one side and a microporous membrane on the other side thereof.
Claim 2
Dependent Claim 30 words
The test device of claim 1 wherein the macroporous membrane contains pores greater than 0.2 µ in diameter and the sucroporous membrane contains pores up to 0.2 µ in diameter.
Claim 3
Dependent Claim 47 words
A diagnostic test device as in claim 1 wherein there is contained in the microporous membrane a colormetric indicator system for the detection of an analyte applied to the macroporous membrane and reagents which are incompatible with the colormetric indicator system are incorporated into the macroporous membrane.
Claim 4
Dependent Claim 28 words
The diagnostic test device of claim 3 wherein the microporous layer contains nicotinamide adenine dinucleotide, glucose-6-phosphate dehydrogenase and diaphorase and the macroporous layer contains hexokinase and a buffer.
Claim 5
Dependent Claim 45 words
A diagnostic device as in claim 1 wherein there is contained in the macroporous layer a material which will inhibit interference with the assay for detection of the analyte in the test sample in the macroporous layer and a chromogenic indicator in the microporous layer.
Claim 6
Dependent Claim 39 words
The device of claim 5 wherein the macroporous layer comprises polyethyleneimine having iodate ion bound thereto to thereby oxidize ascorbate present in the test sample and prevent it from coming in contact with the indicator in the microporous layer.
Claim 7
Dependent Claim 71 words
A diagnostic device for detecting the presence of glucose in a liquid test sample which comprises a permeable polyester support member having on one side thereof a porous polymeric layer containing adenosine triphosphate and hexokinase wherein the pores are greater than 0.2 µ in diameter and on the other side a layer of polyacrylonitrile having impregnated therein a chromogenic indicator together with nicotinamide adenine dinucleotide, glucose-6-phosphate dehydrogenase, diaphorase and a buffer.
Claim 8
Dependent Claim 31 words
A method of detecting the presence and/or amount of glucose in a fluid test sample which method comprises applying the fluid to the macroporous surface of the device of claim 7.
Claim 9
Dependent Claim 62 words
A diagnostic test device for detecting the presence of an analyte in a fluid sample which device comprises a fluid permeable support member having a layer on one side thereof a macroporous membrane comprising polyethyleneimine having impregnated therein iodate ion and on the other side of the support member having a layer of a microporous membrane having dispersed therein a chromogenic indicator.
Claim 10
Dependent Claim 27 words
The device of claim 9 wherein the macroporous membrane comprises a blend of polyethyleneimine, polyacrylonitrile, polyurethane and polyvinyl acetate and the microporous membrane is comprised of polyacrylonitrile.
