Determination of primary structure of protein or peptide
Abstract
Abstract PURPOSE: To determine the primary structure of proteins or peptides, without losing the specimen by washing even in case of a low molecular weight peptide, by reacting the specimen with a polymer having amino groups at the side chains, and then carrying out the Edman reaction. CONSTITUTION: Specimen (e.g. protein having carboxyl groups shown in the figure (a)) is made to react with a polymer having amino acids at the side chain, e.g. the one shown in the figure (b) (e.g. polyornithine having a molecular weight of 100,000W200,000) to obtain a condensate (c). A part of the unreacted amino groups of the polymer are modified with a polar group to obtain a hydrophilic condensate. The condensate is subjected to the ordinary Edman reaction and the terminal amino acids are successively separated and identified. The dissolution-loss of the specimen can be prevented.
