Abstract

Abstract Disclosed is a vaccine which prevents disease caused by Feline leukemia virus (FeLV) and which comprises protease enzyme-inhibited, cell-free, ostensibly virus-free, in vitro produced feline leukemia neoantigens including FOCMA which evoke an immune response in cats by the appearance of antibodies thereto and a FeLV virion gp70 neoantigen which evokes an immune response in cats by the appearance of antibodies thereto.

Total Claims: 16
Claim 1
Independent Claim 62 words
A vaccine for the prevention of disease caused by Feline leukemia virus (FeLV) which comprises protease enzyme-inhibited, cell-free, in vitro produced feline leukemia neoantigens including: (a) a FOCMA-type neoantigen which evokes an immune response in cats by the appearance of antibodies thereto; and (b) a FeLV virion gp70 neoantigen which evokes an immune response in cats by the appearance of antibodies thereto.
Claim 2
Dependent Claim 62 words
A vaccine for the prevention of disease caused by Feline leukemia virus (FeLV) which comprises protease enzyme-inhibited, cell-free, in vitro produced feline leukemia neoantigens including: (a) a FOCMA-type neoantigen which evokes an immune response in cats by the appearance of antibodies thereto; and (b) a FeLV virion gp70 neoantigen which evokes an immune response in cats by the appearance of antibodies thereto.
Claim 3
Dependent Claim 22 words
The vaccine of claim 1 wherein protease enzyme-inhibition is by the addition of an effective proportion of protease inhibitor to said neoantigens.
Claim 4
Dependent Claim 37 words
The vaccine of claim 1 or 2 wherein said neoantigens further include: (a) a FeLV virion p27 antigen; (b) a FeLV virion pl5 neoantigen in precursor form; and (c) a FeLV virion pl2 neoantigen in precursor form.
Claim 5
Dependent Claim 13 words
The vaccine of claim 1 or 2 further including a pharmaceutically acceptable adjuvant.
Claim 6
Independent Claim 83 words
A method for recovering Feline leukemia associated neoantigen from cells infected with Feline leukemia virus, which comprises: (a) culturing said cells in a serum-containing growth medium; (b) transferring and maintaining said cultured cells in a serum-free medium under conditions and for a time adequate to accumulate said neoantigen shed from said cells in said serum-free medium; (c) separating said cells from said neoantigen-rich medium. (d) concentrating at least said neoantigen in said neoantigen-rich medium; and (d) inhibiting protease enzymes in said neoantigen concentrate.
Claim 7
Claim 8
Dependent Claim 12 words
The method of-claim 5 wherein said cultured cells are feline lymphoblastoid cells.
Claim 9
Dependent Claim 15 words
The method of claim 5 wherein said cells are separated in step (c) by centrifugation.
Claim 10
Dependent Claim 24 words
The method of claim 5 wherein said concentration step (d) includes at least one of the following methods: lyophilization, ultrafiltration, or continuous-flow molecular filtration.
Claim 11
Dependent Claim 26 words
The method of claim 5 or 7 wherein said concentration step (d) includes continuous-flow molecular filtration followed by freezing and lyophilization of the resulting neoantigen-containing tenate.
Claim 12
Dependent Claim 20 words
The method of claim 9 wherein said molecular filtration employs a filter having an exclusion size of about 10,000 daltons.
Claim 13
Dependent Claim 10 words
The neoantigen concentrate produced by the method of claim 5.
Claim 14
Dependent Claim 10 words
The neoantigen concentrate produced by the method of claim 10.
Claim 15
Independent Claim 89 words
A method of recovering cell neoantigen associated with virus infected cells wherein said virus exhibits growth characteristics including noncytotoxicity and budding, which comprises: (a) culturing said infected cells in a serum-containing growth medium; (b) transferring and maintaining said cultured cells in a serum-free medium under conditions and for a time adequate to accumulate in said serum-free medium cell neoantigens shed from said cells; (c) separating said infected cells from said neoantigen-rich medium; (d) concentrating at least said neoantigen rich medium; and (e) inhibiting protease enzymes in said neoantigen concentrate.
