Abstract

Abstract Agglutination assays, particularly latex agglutination assays, for simultaneous testing for a multiplicity of ligands. The reagents for use in the assays comprise two or more insoluble colored substances, each substance being adapted to form a distinctively colored agglutinate in the presence of a specific ligand or specific group of ligands.

Claims Analysis

Total Claims: 13
Claim 1
Independent Claim 161 words
A method of testing for the presence of a ligand in an aqueous liquid medium comprising the steps of: A. mixing a sample of the said medium with a reagent comprising particles of a first colour having attached thereto polyclonal antibody bindable tothe said ligand, in admixture with particles of a second colour having attached thereto immunoglobulin from a non-inoculated animal of the same species as that in which the said polyclonal antibody was rasied, whreby non-specific interaction between the said sample and the said reagent is indicated by agglutination of both the said particles of the first colour and the said particles of the second colour and the presence of the said ligand in the said sample, free from said non-specific interaction, is indicated by agglutination of the said particles ofthe first colour alone; B. after an appropriate period of time, inspecting the mixture resulting from step A.; and C. determining the test result from the appearance of said mixture.
Claim 2
Claim 3
Dependent Claim 24 words
The method of claim 1 whrein both the particles of the first colour and the particles of the second colour are of polystryrene latex.
Claim 4
Dependent Claim 14 words
The method of claim 1 wherein the polyclonal antibody was raised in a rabbit.
Claim 5
Dependent Claim 12 words
The method of claim 1 wherein the ligand is a viral antigen.
Claim 6
Dependent Claim 12 words
The method of claim 1 wherein the ligand is a bacterial antigen.
Claim 7
Dependent Claim 15 words
The method of claim 1 wherein the ligand is an antigen characteristic of Haemophilus organisms.
Claim 8
Dependent Claim 15 words
The method of claim 1 wherein the ligand is an antigen characteristic of Neisseria organisms.
Claim 9
Dependent Claim 15 words
The method of claim 1 wherein the ligand is an antigen characteristic of Salmonella organisms.
Claim 10
Dependent Claim 15 words
The method of claim 1 wherein the ligand is an antigen characteristic of Streptococcus organisms.
Claim 11
Dependent Claim 18 words
The method of claim 1 wherein the ligand is an antigen characteristic of Streptococcus organisms of serogroup A.
Claim 12
Dependent Claim 125 words
A test reagent for the presence of a ligand in a sample of an aqueous liquid medium, said reagent comprising particles of a first colour having attached thereto polyclonal antibody bindable to said ligand, in admixture with particles of a second colour having attached thereto immunoglobulin from a non-inoculated animal of the same species as that in which the said polyclonal antibody was raised, whereby non-specific interaction between the said sample and the said reagent is indicated by agglutination of both the said particles of the first colour and the said particles of the second colour and the presence of the said ligand in the said sample, free from said non-specific interaction, is indicated by agglutination of the said particles of the first colour alone.
